Act! Marketing Automation uses the Act! API to pull group data from your database.
If the groups drop-down list is not populated, it is because Act! Marketing Automation is unable to establish a connection to your database through the API. Act! Marketing Automation requires the Act! API to be set-up on your database host, and available by a secure HTTPS endpoint.
Ensure the Act! API has been set up correctly on the database host machine for your deployment type of Act! by following the article below:

If you are still unable to see group data after configuring the Act! API on the database host as above, or you are using a Remote Database that synchronizes with the main database host machine, you may need to update a database preference to point Act! Marketing Automation to the correctly configured API address.
